In the European Union, tests for eye irritation and corrosion are mandatory for the approval of cosmetic products, chemicals, and pharmaceuticals. These tests must comply with the guidelines of the REACH regulation (Registration, Evaluation, Authorization, and Restriction of Chemicals) and the CLP regulation (Classification, Labeling, and Packaging of substances and mixtures).
The Laser Light-Based Opacitometer (LLBO) is a validated device used to measure the opacity of substances in the Bovine Corneal Opacity and Permeability (BCOP) Test Method in accordance with OECD Test Guideline 437. This in vitro method is used to evaluate the potential of substances to cause serious eye damage or irritation.
The OECD Test Guideline 437 describes the BCOP Test Method for Identifying Ocular Corrosives and Severe Irritants, thereby reducing and refining the number of in vivo Rabbit Eye Irritation tests. In 2020, the Test Guideline was updated to allow the use of a Laser Light-Based Opacitometer (LLBO) which was shown to have improved performance over the OP-KIT opacitometer used in the validation of the BCOP test method (https://doi.org/10.1787/9789264203846-en).
